Den näst viktigaste staden i Tjeckien är Brno, som ligger i den sydöstra delen av landet. Det ligger i sammanflödet av floderna Svitava och Svratka i Mährenregionen.
Denna gamla stad bevarar unika historiska byggnader och platser som man bara inte får missa. Den äldsta av dessa är stadshuset Stará Radnice, som är från 1343 och har ett mycket högt torn. I Brno lever industrirevolutionen upp igen i de fabriker från den tiden som bevarats här och som givit staden dess tillnamn Tjeckiens Manchester. Å andra sidan hittar man, bland alla andra sevärdheter i denna historiska stad, Villa Tugendhat, en byggnad från 1929 som är ett utmärkt exempel på den funktionalistiska arkitekturen från mellankrigstiden. Brno var den berömde vetenskapsmannen Gregor Mendels hemstad. Han levde och studerade i det gamla augustinerklostret St. Tomas, en plats man kan besöka. Det är svårt att hitta en like till det rika kulturarvet i staden Brno.
